Th e m icrobiological basic tests of ‘m d t’ inform

p rop erties of m aterials and d isinfectants and h elp
to control m anu factu ring p rocesses and steriliz a-
tion p roced u res.

1 . K E IM Z A H L B E S T IM M U N G
Keimzahlbestimmungen (Bioburden) werden durchge-
führt, um Kenntnis über die mikrobielle Belastung ein-
zelner Medizinprodukte nach oder während der Pro-
duktion zu erlangen.
Da die Wiederfindungsraten von Bakterien und Pilz-
sporen auf einem Medizinprodukt oder dessen Pack-
mittel stark materialabhängig sind, muss vor der eigent-
lichen Keimzahlbestimmung eine Methodenvalidierung
gemäß EN ISO 11737-1 durchgeführt werden. Die
materialabhängige Wiederfindungsrate wird dann zur
Korrektur der gefundenen Keimbelastungswerte heran-
gezogen und führt zu einer realitätsnahen Ermittlung
der zu erwartenden Keimbelastung. Bei der Validierung
der Bioburdenbestimmung können zwei unterschied-
liche Richtlinien herangezogen werden. Einerseits kann
die EN ISO 11737-1 herangezogen werden. Die Norm
schreibt den Einsatz von den folgenden drei Prüfkeimen
vor: Staphylococcus aureus, Fusarium solani und Bacil-
lus subtilis. Als Alternative kann auch die Validierung
nach den harmonisierten Pharmakopöen (Ph.Eur., USP
und JP) durchgeführt werden. Laut diesen harmonisier-
ten Pharmakopöen sind 5 Prüfkeime einzusetzen: Sta-
phylococcus aureus, Pseudomonas aeruginosa, Bacillus
subtilis, Candida albicans und Aspergillus brasiliensis.

1. BIOBURDEN DETERMINATIONS
The bioburden of a product is determined in order to
assess the number of viable microorganisms present on
a medical device or its packaging material during or at
the end of the manufacturing process.
As the recovery rates of bacteria and molds, which
adhere to a medical device or its package, strong-
ly depend on the respective material properties, any
bioburden determination procedure needs to be vali-
dated according to EN ISO 11737-1. The determined
recovery rate is then applied to the measured biobur-
den value in order to evaluate the real microbiological
status of a product. For the validation of the biobur-
den determination two different guidelines can be used:
First, EN ISO 11737-1 can be used. This standard
specifies the use of the following three microorganisms:
Staphylococcus aureus, Fusarium solani and Bacillus
subtilis. Alternatively, the validation can also be per-
formed using the harmonized Pharmacopeia (Ph. Eur.,
USP, and JP). According to these three harmonized
Pharmacopeia a total of five microorganisms must be
used: Staphylococcus aureus, Pseudomonas aerugino-
sa, Bacillus subtilis, Candida albicans and Aspergillus
brasiliensis.

Die Prüfung auf Sterilität weist nach, ob sich auf einem
geprüften Produkt vermehrungsfähige Organismen
befinden. Sie dient jedoch nicht zum Nachweis eines
Sterilitätserfolges nach Sterilisation. Dazu ist eine Vali-
dierung des jeweiligen Sterilisationsverfahrens erfor-

Sterility tests confirm that a product does not contain
any viable microorganisms. Sterility tests, however,

Einige Produkte werden vom Patienten mehrfach ver-
wendet oder über einen längeren Z eitraum aufge-
braucht. In diesen Fällen sind zuverlässige Desinfektion
oder Konservierung des Produktes erforderlich. Nach-
folgend sind beispielhaft Prüfungen auf ausreichende
antimikrobielle Wirkung dargestellt.

Some medical devices can be applied repeatedly or can
be used up by patients over a longer period of time. For
such products effective disinfectants, respectively effec-
tive preservative agents, are req uired. In the following,
some tests are presented which are applied to investi-
gate such antimicrobial properties.

Produkte, die sterilisiert werden sollen, müssen gemäß
MDD unter angemessenen und überwachten Bedin-
gungen hergestellt sein. Die Europäische G MP-Richt-
linie gibt hierzu hilfreiche Anregungen, wie die Ü ber-
wachung des Produktionsbereiches und der darin
arbeitenden Mitarbeiter realisiert werden kann.
In Z usammenarbeit mit dem Auftraggeber erarbei-
tet die ‘mdt’ ein angemessenes Monitoringprotokoll,
welches Z eitpunkte und Positionen der Probennahme
festlegt und G renz- und Alarmwerte definiert. Auf die-
se Weise wird der mikrobiologische Status der Produk-
tionsumgebung anhand der folgenden Testsysteme fest-
gehalten und bewertet:
– Oberflächenkeime: Abklatschtests
(z. B. CASO-Agar Platten mit Enthemmern)
– Luftkeime: Sedimentationstests
(z. B. CASO-, SAB-Agar Platten)
– Wasserkeime: (z. B. CASO-, SAB-Agar Platten)
Darüber hinaus bietet ‘mdt’ Trainingskurse zur Pro-
duktions-, Betriebs- und Personalhygiene an.

5 . MIC ROBIOL OG IC AL MONITORING OF
P RODUC TION
According to the M D D devices which are supposed to
be sterilized must be manufactured under appropriate
and controlled conditions. In this regard the European
G M P G uideline offers helpful suggestions as how the
practical performance concerning the production area,
and the personnel working in this area, can be realized.
In cooperation with the manufacturer, ‘mdt’ develops
a suitable sampling plan which specifies the time regi-
men and sampling positions in the production area
and which determines action and alert limits for each
sampling position. Thereby the microbiological status
of the controlled environment can be documented and
evaluated for the following test systems:
– M icroorganisms on surfaces: Total colony count
(e. g., Tryptic Soy Agar with Neutralizer)
– Airborne microorganisms: Sedimentation test
(e. g., CASO-, SAB-Agar Plates)
– M icroorganisms in water supplies: Plating of
samples (e. g., CASO-, SAB-Agar Plates)
Furthermore, ‘mdt’ offers hygiene training courses for
employees working in a cleanroom environment.
