Flusskrebs Neuromuskulre Endplatte
basischsauer
NH4Cl (60mMol) added, goes into neuron, Na/H+ Antiporter begins to work, wash out, acidic intracellularIncrease in amplitude after acidificationLow Na due to exchange NH4Cl (to keep osmolarity)Bei basisch EPSP noch gering, wenn sauer geht stark hoch, beruhigt sich dann aber auch wiederRP: normal, bei Alkalisierung Depolarisation, bei Acidifizierung Repolarisation17
Flusskrebs Neuromuskulre Endplatte
basischsauer
After washout quick recovery in EPSP (after few minutes (3-4))Slow recovery concerning the pH (30)In Salzlsung als Referenz, dann mit CO2 deutliche Abnahme (s. auch Folie 12, Nulllinie), kann ausgewaschen werdenZweite Graphik mit pH Imaging (an Wright State University, OH):Grn fr normalen pH, zu Beginn in Salzlsung normal, dann mit CO2 gering, kann ausgewaschen werdenAllerdings Hhe der Amplitude schnell wieder regenerierbar durch CO2 Auswaschung, aber pH nur langsam auf ursprngliches Niveau

NH4Cl60mmol/lCO2+NH4Cl
Measurement EPSP after stimulationSaline normal, as referenceWith CO2 no response; CO2 and glutamate no response also (no depolarization even if transmitter added!)Recovery after washoutDepolarization with low pH (but not with CO2); but no response with glutamate Recovery after washout
Mit Glutamat Desensibilisierung, erst EPSP (starke Depolarisation), dann nicht mehr, auch mit saurem pHWenn CO2 dazu, dann gar nichtsSieht hier leider etwas verzerrt aus, weil gar keine Antwort.
But add glutamate in saline ...rapid depolarization then back to resting membrane potential due to the glutamate receptors desensitizing and not being open while being desensitized.
However with CO2 and glutamate ...no depolarization... Maybe that is why EPSPs not observed...
19Drosophila Neuromuskulre Endplatte
basischsauer
Same as Crayfish: when basic then depolarization, when acidic lowering RPDecrease in apmplitude of EPSP upon alkalizationJumps up when acidification occurs, but then decreasesHier genau gleich, wie eben am Flusskrebs erklrt:Bei Alkalisierung RP depolarisiert, dann bei Ansuerung wieder Repolarisation, nicht auf ursprnglichen WertDann bei EPSP Amplitude: hier geringer bei Alkalisierung (nicht so Flusskrebs), aber bei Ansuerung gleiches Bild: erhhte AmplitudepH Messung wie Flusskrebs; erholt sich nur langsam 20Drosophila Neuromuskulre Endplatte
RP angeschautLow RP with pH 7.2 (physiological)When pH reduced to 5.0 slight depolarization, when glutamate added strong depolarization (physiological, thats the way it should work)Again back to low RP upon recoveryWhen CO2 added slight depolarization, but clearly not as strong; when glutamate added no depolarization somehow blocks the glutamate receptorsSame thing shown in second graph (might leave it out, only have half an hour to present)Also was hier interessant ist: CO2 mit anderer Wirkung auf RP als pH; wenn prsynaptisch, dann drfte die Sache mit dem Glutamat bei beiden nicht funktionieren, also muss postsynaptisch arbeiten21
Drosophila Neuromuskulre Endplatte
basischsauerpH 7.2Co2 / NH4ClpH 7.2
No EPSP after exposure to CO2, but recovery possibleIf pH lowered EPSP decreased, but still there; recovery possible22Propionsure Drosophila

23Drosophila: Minis
No stimulation just spontaneous minis (fly muscle)... saline, add Cd2+ (blocks all volt gated channels on surface of pre synaptic terminal) add P Acid with Cd2+ increase in minis... THUS Ca2+ not coming from Voltage gated Ca channels in plasma membrane... it is being dumped from ER within the Presynaptic terminal.24ZusammenfassungCO2 hat nicht nur einen ansuernden EffektVerschiedene Wirkungsmechanismen von CO2 und pHBlockieren der Neurotransmission durch CO2Direkte Wirkung CO2 auf postsynaptische ZelleDepolarisation RP durch Ansuern Erhhung EPSP durch Ansuern (Flusskrebs)Propionsure erhht ErregbarkeitFreisetzung Ca2+ aus Intrazellularraum??
