Peroxisomen
Sind von einer einzigen Membran umgeben.
0.2-1.5µm Durchmesser.
“Peroxisom” ist auf die H2O2 produzierende Enzymaktivität, welche in Peroxisomen zu finden ist, zurückzuführen.
Der Ursprung der P ist unklar.
Ps bsitzen kein Genom.
Spezialisierte Zellen besitzen bis zu 300Ps
Funktion von Peroxisomen
Reduktion von O2 zu H2O2 durch OxidasenDurch Catalasen wird H2O2 in H2O und 1/2 O2 zerlegtAlcohol-Oxidase, Urat Oxidase, D-Aminosäure Oxidase….
ß-Oxidation von Fettsäuren (in Pflanzen und Pilzen findet die Verwertung von Fettsäuren ausschließlich in Peroxisomen statt).
In Pflanzenzellen keimender Samen: der Glyoxylat Zyklus ermöglicht die Bildung von Succinylsäure aus Acetyl CoA (aus dem Fettsäurezyclus) Succinylsäure kann direkt zur Bildung von Glukose verwendet werden (Gluconeogenese).
Peroxisomen
Entstehen durch Teilung von existierenden Peroxisomen.
Da Peroxisomen kein Genom besitzen, müssen alle Proteine importiert werden.
Proteine werden durch Signalsequenzen erkannt und importiert.In der Regel ist ein kurzes, C-terminales Signalpeptid vorhanden.Proteine werden in Peroxisomen bereits im gefalteten Zustand importiert. Der Importmechanismus ist schlecht verstanden.
Patienten mit Zellweger Syndrom besitzen leere Peroxisomen. Dies führt zu Leber, Nieren und Nervenschädigung und schließlich zum Tod.
